--- res: bibo_abstract: - Clathrin-mediated endocytosis (CME) and its core endocytic machinery are evolutionarily conserved across all eukaryotes. In mammals, the heterotetrameric adaptor protein complex-2 (AP-2) sorts plasma membrane (PM) cargoes into vesicles through the recognition of motifs based on tyrosine or di-leucine in their cytoplasmic tails. However, in plants, very little is known on how PM proteins are sorted for CME and whether similar motifs are required. In Arabidopsis thaliana, the brassinosteroid (BR) receptor, BR INSENSITIVE1 (BRI1), undergoes endocytosis that depends on clathrin and AP-2. Here we demonstrate that BRI1 binds directly to the medium AP-2 subunit, AP2M. The cytoplasmic domain of BRI1 contains five putative canonical surface-exposed tyrosine-based endocytic motifs. The tyrosine-to-phenylalanine substitution in Y898KAI reduced BRI1 internalization without affecting its kinase activity. Consistently, plants carrying the BRI1Y898F mutation were hypersensitive to BRs. Our study demonstrates that AP-2-dependent internalization of PM proteins via the recognition of functional tyrosine motifs also operates in plants.@eng bibo_authorlist: - foaf_Person: foaf_givenName: D foaf_name: Liu, D foaf_surname: Liu - foaf_Person: foaf_givenName: R foaf_name: Kumar, R foaf_surname: Kumar - foaf_Person: foaf_givenName: Claus foaf_name: LAN, Claus foaf_surname: LAN - foaf_Person: foaf_givenName: Alexander J foaf_name: Johnson, Alexander J foaf_surname: Johnson foaf_workInfoHomepage: http://www.librecat.org/personId=46A62C3A-F248-11E8-B48F-1D18A9856A87 orcid: 0000-0002-2739-8843 - foaf_Person: foaf_givenName: W foaf_name: Siao, W foaf_surname: Siao - foaf_Person: foaf_givenName: I foaf_name: Vanhoutte, I foaf_surname: Vanhoutte - foaf_Person: foaf_givenName: P foaf_name: Wang, P foaf_surname: Wang - foaf_Person: foaf_givenName: KW foaf_name: Bender, KW foaf_surname: Bender - foaf_Person: foaf_givenName: K foaf_name: Yperman, K foaf_surname: Yperman - foaf_Person: foaf_givenName: S foaf_name: Martins, S foaf_surname: Martins - foaf_Person: foaf_givenName: X foaf_name: Zhao, X foaf_surname: Zhao - foaf_Person: foaf_givenName: G foaf_name: Vert, G foaf_surname: Vert - foaf_Person: foaf_givenName: D foaf_name: Van Damme, D foaf_surname: Van Damme - foaf_Person: foaf_givenName: Jiří foaf_name: Friml, Jiří foaf_surname: Friml foaf_workInfoHomepage: http://www.librecat.org/personId=4159519E-F248-11E8-B48F-1D18A9856A87 orcid: 0000-0002-8302-7596 - foaf_Person: foaf_givenName: E foaf_name: Russinova, E foaf_surname: Russinova bibo_doi: 10.1105/tpc.20.00384 bibo_issue: '11' bibo_volume: 32 dct_date: 2020^xs_gYear dct_identifier: - UT:000600226800021 dct_isPartOf: - http://id.crossref.org/issn/1040-4651 - http://id.crossref.org/issn/1532-298x dct_language: eng dct_publisher: American Society of Plant Biologists@ dct_title: Endocytosis of BRASSINOSTEROID INSENSITIVE1 is partly driven by a canonical tyrosine-based Motif@ fabio_hasPubmedId: '32958564' ...
